Lab projects in Salmonella

Interaction of bacterial effectors with targets in epithelial cells, macrophages and lymphocytes.

We are currently examining effectors of the SPI-2 system (see Figure 3 right) for their effects on sterol recruitment, metabolism and bacterial virulence in the mouse model.

Recruitment of host sterol by S. Typhimurium.

Up to 30-40% of total host cholesterol is diverted to the SCV and our efforts focus on its source and mechanisms of trafficking to the SCV (see Figure 4 right).

Perturbation of the host sterol metabolism.

We are investigating the requirement for non-sterol precursors in protecting infected cells from apoptotic and/or necrotic death.

Salmonella microarrays and other gene screens.

These are employed to identify subsets of S. typhimurium virulence determinants required for intracellular bacterial replication (see Figure 5 right).
